He is best known for his television roles, including Thomas Cromwell in the historical drama series The Tudors (2007–2009), Franklin Mott in True Blood (2010), and Sarek in Star Trek: Discovery (2017–2019). Frain has also appeared in other notable TV shows such as The White Queen (2013), The Tunnel (2013), Orphan Black (2015–2017), and Gotham (2015–2016).
